The ProtoPrep II System
provides a meltable synthetic gel matrix as an
acrylamide replacement for the separation, purification,
and recovery of proteins. After the gel is run, proteins
from 10 to 220kD can be located, excised, and then
recovered from the matrix by melting the gel section.
The ProtoPrep II gel will melt only once and then
remains liquefied. The protein of interest may be
processed directly in the aqueous phase or further
purified using the methods provided.
ProtoPrep II can be run in any standard vertical
electrophoresis apparatus. Simply combine the ProtoPrep
II concentrate, ProtoPrep II Buffer, and water, initiate
and cast the gel. Gels are ready to use in 1 hour.
For
maximum protein recovery,
Insite, National Diagnostics¹ run-time protein
visualization system is recommended for use with
ProtoPrep II. Insite incorporates a unique dye/buffer
combination which stains proteins during the gel run.
Bands containing as little as 10 ng of protein can be
detected with just a 30 minute post-run destaining in
water.
ProtoPrep II is supplied in 450 ml kits containing 200ml
of ProtoPrep II Concentrate, 200ml of ProtoPrep II Gel
Buffer and 50 ml of ProtoPrep II Dissolution Reagent.
The buffers
accompanying the ProtoPrep II kit are designed for
protein electrophoresis. To carry out electrophoresis of
DNA or RNA, please separately order ProtoPrep II
Concentrate, ProtoPrep II Dissolution Reagent, and TTE
Glycerol Tolerant Buffer.
Storage: Recommended storage of ProtoPrep II Concentrate
is at 4-8oC for up to one year.

AcrylaGel is a
ready-to-use, 30% acrylamide solution in
distilled/deionized water. Because acrylamide in its
crystalline form is subject to self-polymerization, the
purity and consistency of polymerized AcrylaGel is
superior to that obtainable with powdered acrylamide.
AcrylaGel can be crosslinked with Bis-AcrylaGel, a
ready-to-use, 2% solution of methylene bisacrylamide
with the same advantages as the AcrylaGel acrylamide
solution. However, any powdered crosslinking reagent can
be used with AcrylaGel.
AcrylaGel contains 300 grams of acrylamide per liter of
solution. Bis-AcrylaGel contains 20 grams of methylene
bisacrylamide per liter of solution.
Storage: AcrylaGel and Bis-AcylaGel are stable for 24
months when stored tightly capped in a dark area at room
temperature (20oC).

National Diagnostics
ProtoGel forms an electrophoresis matrix that is ideal
for the separation of proteins and polypeptides.
ProtoGel is a stabilized, ready-to-use 30% (w/v)
acrylamide/methylene bisacrylamide solution (37.5:1
ratio), manufactured from the highest quality materials,
from which virtually all impurities have been removed.
ProtoGel has zero acrylic acid content, eliminating the
fixed charges that cause band streaking. Additionally,
oxidation products such as aldehydes have been removed
by a selective adsorption process. Aldehydes can attack
proteins, altering the structure of the sample, and thus
altering Rf values. With ProtoGel, you can trust that
your results will be consistent one electrophoretic run
to the next.
Storage: ProtoGel is stable for 24 months when stored
tightly capped in a dark area at room temperature (20oC).
